Quantitative ratio of mRNA expression of IGF/INS system receptors in multiple myeloma

چکیده

Background. Individuals with increased expression of components the IGF/INS system, are more likely to develop various malignancies. And in case when system overexpressed tumors, this adversely affects prognosis disease, including leading a decrease relapse-free survival. A characteristic feature is ability same ligands bind different receptors and vice versa (cross interactions) activate signaling pathways cell. This requires an integrated approach study its components, namely, quantitative ratio individual components. The result obtained will make it possible determine combinations ligand-receptor bonds and, ultimately, have both prognostic evaluative value: terms therapeutic target. Aim. To establish mRNA receptors: IR-A, IR-B, IGF-1R, IGF-2R IM9 lymphoblastoid cell line three myeloma lines: RPMI1640, RPMI8226, H929, identify frequency these mononuclear fraction bone marrow aspirates from treated patients multiple myeloma. Materials methods. We used human cells 3 types cells, differing degree differentiation as well 19 stage III Expression was studied by real-time reverse transcription polymerase chain reaction aspirate samples semi-quantitative reaction. Results. During study, we found that within each line, receptor IR-A predominant compared IR-B . Patients MM high minimum IGF-1R:IR-A IGF-1R:IR-B for ratios high. IGF-2R:IR-A maximum or times less. Conclusion. Based on mRNA, state there probability presence IGF-1R/IGF-1R IR-A/IR-A homodimers, IGF-1R/IR-A heterodimer. These data value, since suggest significant increase mitogenic effect due activation ligands: IGF-1, IGF-2 INS, which unfavorable factor, especially patient concomitant Diabetes mellitus prescribed insulin therapy along chemotherapy. our findings, recommend simultaneously inhibiting IGF-1R

Correlation of Adiponectin mRNA Abundance and Its Receptors with Quantitative Parameters of Sperm Motility in Rams

Objective Adiponectin and its receptors (AdipoR1 and AdipoR2), known as adiponectin system, have some proven roles in the fat and glucose metabolisms. Several studies have shown that adiponectin can be considered as a candidate in linking metabolism to testicular function. In this regard, we evaluated the correlation between sperm mRNA abundance of adiponectin and its receptors, with sperm moti...

mRNA Expression of VEGF and Its Receptors in Fallopian Tubes of Women with Ectopic Pregnancies

Background Establishment of viable pregnancy requires embryo implantation and placentation. Ectopic pregnancy (EP) is a pregnancy complication which occurs when an embryo implants outside of the uterine cavity, most often in a fallopian tube. On the other hand, an important aspect of successful implantation is angiogenesis. Vascular endothelial growth factor (VEGF) is a potent angiogenic factor...

Methylation and mRNA expression levels of P15, death-associated protein kinase, and suppressor of cytokine signaling-1 genes in multiple myeloma

Objective(s): The aim of this study was to investigate the methylation status and mRNA expression levels of P15, death-associated protein kinase (DAPK), and suppressor of cytokine signaling-1 (SOCS1) genes in multiple myeloma (MM). Materials and Methods: The bone marrow samples of 54 MM patients were collected and the methylation status of the P15, DAPK, and SOCS1 gene promoter regions was dete...
